You can defrag individual drives in Vista SP1
After installing Windows Vista Service Pack 1, you’ll note a change in the Disk Defragmenter GUI. There is a new button added that allows you select the drives that you want to defragment. Note that in Windows Vista pre-SP1, you can use the command-line defrag tool (defrag.exe) to defragment individual drives.

When you install Windows Vista Service Pack 1, it removes the Search… option from the context menu of drives and file system folders. However, the missing option can be restored using a registry edit.
